چکیده

The structural and dynamic organization and basic physicochemical features of 1,2dimyristoyl-sn-glycero-3-phosphocholine (DMPC) lipid bilayers were characterized upon the presence of two OxPLs, 1-palmitoyl-2-(9’-oxo-nonanoyl)-sn-glycero-3-phosphocholine (PoxnoPC) and 1-palmitoyl-2-glutaryyl-sn-glycero-3-phosphocholine (PGPC). The hydration behaviour of these systems was also studied by using the isotope effect of heavy water. The characterization of these systems was therefore performed by a combined biophysical approach of Differential Scanning Calorimetry (DSC) and Solid State 31 P, 2 H Nuclear Magnetic Resonance (NMR) to unravel in atomic detail the complex interactions between different types of lipids and the impact of water on these lipid bilayers.

Oxidized phosphatidylcholines facilitate phospholipid flip-flop in liposomes.

Lipid asymmetry is a ubiquitous property of the lipid bilayers in cellular membranes and its maintenance and loss play important roles in cell physiology, such as blood coagulation and apoptosis. The resulting exposure of phosphatidylserine on the outer surface of the plasma membrane has been suggested to be caused by a specific membrane enzyme, scramblase, which catalyzes phospholipid flip-flo...

The lipid whisker model of the structure of oxidized cell membranes.

An essential feature of the innate immune system is maintaining cellular homeostasis by identifying and removing senescent and apoptotic cells and modified lipoproteins. Identification is achieved through the recognition of molecular patterns, including structurally distinct oxidized phospholipids, on target cells by macrophage receptors. Both the structural nature of the molecular patterns rec...

Lipid Bilayers Are Long-Lived on Solvent Cleaned Plasma-Oxidized poly(dimethyl)siloxane (ox-PDMS)

Although it is well known that phospholipids self-assemble on hydrophilic plasma-oxidized PMDS surfaces (ox-PDMS) to form cell membrane mimetic bilayers, the temporal stability of phospholipid membranes on these surfaces is unknown. Here we report that phospholipid bilayers remain stable on solvent-cleaned ox-PDMS for at least 132 hours after preparation. Absent solvent cleaning, the bilayers w...

Self-healing Biological Molecules for Use in Engineering Materials

Phospholipid molecules are one of the fundamental building blocks of cell membranes in living organisms. These molecules are amphipathic with two hydrophobic fatty acid chains (tails) linked to a phosphate containing hydrophilic group (head) by a glycerol molecule. A bilayer lipid membrane (BLM), which is two phospholipids thick, is a 6-10 nm thick membrane and can be used to seal a porous mate...

Surface Recognition and Complexations Between Synthetic Poly(ribo)nucleotides and Neutral Phospholipids and Their Implications in Lipofection

Thermodynamic features related to preparation and use of self-assemblies formed between multilamellar and unilamellar zwitterionic liposomes and polynucleotides with various conformation and sizes are presented. The divalent metal cation or surfactant-induced adsorption, aggregation and adhesion between single- and double-stranded polyribonucleotides and phosphatidylcholine vesicles was followe...
